Wright had won the opening leg, but Littler was quickly level as he won the second.
The Scot hit back in the third with two double tops in order to lead the match 2-1.
Littler persisted and dominated the fourth leg by hitting a key 180 before finishing with a double 10.
The darts wonderkid completed the win as he wrapped it up 6-4.
The result means that Littler will finish at the top of the Premier League Darts table at his first-ever attempt.
Michael van Gerwen and Luke Humphries have already qualified for the play-offs at the O2 Arena in London on May 23.
The second match saw a straight shootout between Michael Smith and Nathan Aspinall for the final qualifying spot.
The former world champion managed to come out on top, leapfrog Aspinall into the fourth, and book his spot in London.
